摘要猪屎豆为豆科野百合属植物,广泛分布在我国南方各省。其中所含的各类生物碱有很大的药用价值,很多已经用于临床治疗。
以海南岛产的猪屎豆种子为原料,采用无水乙醇提取法,用石油醚,氯仿多次除杂,最后用混合溶剂重结晶得到有效成分,并用核磁共振氢谱、碳谱以及质谱对其进行结构表征,以及它对人胃癌细胞MGC-803、人肝癌细胞HepG-2、人乳腺癌细胞MCF-7体外增殖影响的研究。结果表明:用核磁共振法证实有效成分为光萼猪屎豆碱,光萼猪屎豆碱对人胃癌细胞MGC-803、人肝癌细胞HepG-2、人乳腺癌细胞MCF-7体外增殖有抑制作用,细胞凋亡较明显,且在测定范围内随浓度增大,抑制作用越强。28231

Title Extraction and Separation of Usaramine in Crotalaria and the tumor cell Killing effect in vitro
Abstract
Crotalaria is leguminous crotalaria plants, distributed in the south of china widely. Various types of alkaloids that crotalaria contained have great medicinal value, many of them have been used in clinical treatment.
The raw material is crotalaria seed produced in Hainan Island, extracted it by ethanol method, using petroleum ether, chloroform to remove its impurity repeatedly, and finally, using mixed solvent recrystallized to obtain effective components. And characterized its structure by NMR hydrogen spectrum, carbon spectrum and mass spectrum, as well as the study on the proliferation effect of human gastric cancer cell MGC-803, human hepatoma cell HepG-2, and human breast cancer cell MCF-7. The results show that the active ingredient that has been confirmed by nuclear magnetic resonance spectroscopy is porella crotalaria base, porella crotalaria base has inhibition on the proliferation of human gastric cancer cell MGC-803, human hepatoma cell HepG-2 and human breast cancer cell MCF-7, apoptosis is obvious, and in the measurement range with the increase of concentration, the inhibition is stronger.
