摘要:为深入了解抗除草剂转基因油菜的基因向野芥菜的渗入,以抗草甘膦和抗草丁膦转基因油菜与野芥菜的回交1代子5代(BC1F6)、回交2代子4代(BC2F5)、回交3代子3代(BC3F4)为实验材料,采用SSR检测方法,分析了16对C-染色体特异片段在后代中的遗传规律。实验结果表明,16对C-染色体组特异性引物编码的基因片段能以不同频率向回交后代传递。其中16对C-染色体组特异性引物在正反交携带草甘膦和草丁膦基因的BC1F6中每条引物平均被检测到的频率是4.40%、4.84%和16.72%和13.75%;在正反交携带草甘膦和草丁膦基因的BC2F5中每条引物平均被检测的频率分别为5.94%、10.78%和 19.69%、19.22%。在正反交携带草甘膦和草丁膦基因的BC3F4中每条引物平均被检测的频率分别为11.09%、14.69%和 17.97%、18.13%。综上所述,抗草丁膦转基因油菜的C-染色体组特异片段向野芥菜的传递频率要比抗草甘膦转基因油菜传递频率高。以上研究结果将为抗除草剂转基因油菜的安全性评估的深入开展提供依据。25642 毕业论文关键词:抗草甘膦转基因油菜,抗草丁膦转基因油菜,野芥菜,C-染色体组,SSR
Introgression of specific fragment of C – chromosome from transgenic Brassica napus L. into its backcross progenies (BC1F6,BC2F5,BC3F4) between transgenic oilseed rape and wild B. juncea

Abstract:In order to understand introgression in two herbicide resistant transgenic oilseed rape (B. napus) to wild Brassica juncea, we used BC1F6、BC2F5 and BC3F4 (the backcross progenies from wild Brassica juncea and F1 hybrid of Brassica juncea×transgenic glyphosate- or glufosinate-tolerant oilseed rape) as experimental materials. The persistence of C-chromosome fragments B. napus in backcross progenies were examined by specific simple sequence repeat (SSR) markers. The results showed that C-chromosome fragments coded by 16 pair primers transferred with different frequency in inpidual backcross plants. The average frequency of detected primers were 4.40% and 4.84% in BC1mF6R and BC1pF6R, and 16.72% and 13.75% in BC1mF6L and BC1pF6L, respectively. The average frequency of detected primers were 5.94% and 10.78% in BC2mF5R and BC2pF5R, and 19.69% and 19.22% in BC2mF5L and BC2pF5L, respectively. The average frequency of detected primers were 11.09% and 14.69% in BC3mF4R and BC3pF4R and 17.97% and 18.13% in BC3mF4L and BC3pF4L, respectively. These results indicated that the inheritance of fragment of C–chromosome in the same backcross progenies was much more from transgenic glufosinate-tolerant oilseed rape than from transgenic glyphosate-tolerant oilseed rape. The above results will provide more experiment basis for the safety assessment of genetically modified oilseed rapeseed.
