摘要:磷酸盐的去除是污水处理的关键步骤。细菌细胞表面展示技术的应用,可将磷酸盐吸附蛋白表达于细胞表面,从而克服细胞对磷摄入量的限制,做到高效除磷。本研究通过设计合适的引物,用PCR的方法扩增得到大肠杆菌磷酸盐吸附蛋白编码基因phoS,将inaQ-N与phoS融合并插入相应大肠杆菌表达载体,构建inaQ-N-phoS大肠杆菌细胞表面展示体系,即以InaQ-N为载体蛋白将具有磷酸盐吸附能力的磷酸盐吸附蛋白PBP展示于大肠杆菌细胞表面。结果表明,构建的细胞表面展示系统能有效吸附溶液中的磷酸盐。6896

Functional cell surface display of phosphate binding protein using Ice nucleation protein
Abstract: Phosphate removal is a key step in wastewater treatment. The phsophate binding protein (PBP) can be displayed on the cell surface by the construction of bacterial cell surface display system. This method can overcome the limitation of phosphate intake by cells. In this study, a phosphate binding protein encoding gene named phoS from Escherichia coli was cloned by PCR. Then, the inaQ-N and phoS gene was fused and inserted into the plasmid. The PBP can be displayed on the bacterial surface using InaQ-N as carrier protein. The results in this study showed that the concentration of phosphate was reduced by biosorption of this cell surface display system.
