摘要赤拟谷盗是一种重要的世界性储粮害虫,其幼虫和成虫均可危害面粉等粮作物,且成虫体表的臭腺可以分泌含苯醌等致癌物质的臭液,使面粉等被害物结块、变色、发臭而不能食用,造成巨大的经济损失。复眼是赤拟谷盗的主要视觉器官,由许多小眼组成,在感知外界、觅食、求偶和避敌等方面都起着非常重要的作用。视蛋白是存在于小眼内光感受细胞膜的受体,经过长期的进化和适应,赤拟谷盗的视蛋白进化为长波(LW)视蛋白和紫外线敏感(UV)视蛋白两类。为了明确赤拟谷盗UV视蛋白基因的分子进化,本研究利用RACE技术克隆得到赤拟谷盗UV视蛋白基因的cDNA序列全长,然后和其它昆虫已提交的视蛋白基因序列进行比对以及同源性分析,构建相应的系统发育树。结果显示,克隆得到的赤拟谷盗UV视蛋白基因的cDNA序列全长为1256bp,其编码蛋白含有333个氨基酸。根据赤拟谷盗和杂拟谷盗UV基因序列比对结果,共发现15个变异位点。系统发育树表明赤拟谷盗与其他一些鞘翅目昆虫UV视蛋白基因的同源性较高,其中赤拟谷盗和杂拟谷盗聚为一支,显示它们的种间亲缘关系最近。赤拟谷盗视蛋白基因的克隆与序列分析进化,为深入研究视蛋白在储粮害虫趋光行为中所发挥的作用奠定了基础,不仅在理论上具有重要的意义,在实际的应用中也有潜在价值,可能为储粮害虫的生物绿色防治提供新的思路和方法。72216

Molecular Cloning and Sequence Analysis of UV Opsin in Tribolium castaneum

Abstract Tribolium castaneum is an important world stored grain pest。 Both larvae and adults can harm flour, wheat bran, corn, rice and cereals, and also adult body scent glands can produce benzoquinone carcinogenic substances such as smelly liquid secretion。 The polluted flour and other victims were agglomerate and smell, and they could not be eaten, causing a huge economic loss to agricultural production。 Ommateum is the main organ of vision, composed of many small eyes, and it plays an important role in the perception of the outside world, feeding, courtship, learning and memory, etc。 Opsin is photoreceptor cell membrane receptor in small eyes, and after long-term light adaptation, the visual pigments of T。 castaneum evolve into two categories: long-wavelenth (LW) opsin and ultra-violet (UV) opsin。 In this study, to clarify the molecular evolution of UV opsin in T。 castaneum, the cDNA sequence of UV opsin in T。 castaneum was cloned by rapid amplification of cDNA ends ( RACE), the sequences were aligned, and phylogenetic tree was constructed。 The results showed that UV opsin cDNA sequence is 1256 bp in length, which encodes a polypeptide of 333 amino acids residues。 There were 15 mutation sites in UV opsin genes between species。 Phylogeny incorporating other species revealed that, UV opsin genes of T。 castaneum and T。 confusum were highly homologous, and T。 castaneum were more closely related to T。 confusum。 The molecular cloning of UV-opsin gene and sequence analysis lay the foundation for further study on the role of opsin in phototaxis behavior of grain pests。 It not only has important significance in theory, there are also potential value in practical applications and may provide new ideas and methods for the prevention and elimination of pests in stored grain。
