摘要本研究以半夏(Pinellia ternata)为实验材料,对从其植株内分离得到的内生细菌进行固氮能力、解磷能力、产IAA活性检测,筛选出具有潜在促生活性的菌株。并进行小麦促生筛选试验作为初步筛选,探究半夏内生菌对小麦幼苗生长的影响,为研究半夏内生菌与半夏植株生长发育的相互关系提供一定的理论依据,最终指导半夏药材的优质高产。本研究共对6株半夏内生细菌PtB6,PtB13,PtB35,PtB36,PtB47,PtB49进行促生指标筛选,以无氮培养基接种法进行菌株固氮活性检测,以透明圈法进行菌株解磷能力检测,以Salkowski比色法进行菌株产IAA测定。结果发现该6株菌均至少存在一项潜在促生指标活性,其中PtB6、PtB35、PtB36、PtB49等4株菌可以在无氮培养基上正常生长;PtB6及PtB49两株菌有解无机磷活性,该6株菌中未筛选到能够溶解有机磷的菌株;除PtB6外,其余5株菌均检出有不同程度的产生IAA能力,以PtB35产生IAA能力最强,其发酵液IAA含量为14。05±1。72μg/mL。PtB49兼具固氮活性、溶解无机磷能力及产IAA能力,在小麦促生筛选试验中,PtB49显示出最强的促生能力。72220

The Influence of Endophyte Isolated from Pinellia ternata on the Growth of Wheat Seeding

Abstract

This study used Pinellia ternata as experimental material and gave nitrogen fixation ability,phosphate solubilizing ability and IAA producing ability tests to bacteria isolated from them and inquired into their influence on the growth of wheat seeding,then screened out the strains who have growth-promoting potential and studied their relationship with the growth and development of Pinellia ternata plants。Ultimately the result can be the scientific basis of high quality, high yield for Pinellia ternata 。Six strains tested (PtB6,PtB13,PtB35,PtB36,PtB47,PtB49) are stored in the Key Laboratory of Biotechnology for Medicinal Plant of Jiangsu Province。 This study used nitrogen-free medium inoculation to do nitrogen fixation ability testing,Salkowski colorimetric method to do IAA production testing,and used transparent ring method to assess phosphate solubilizing ability 。The result showed that all these six stains at least have one activity indicator of growth-promoting potential。PtB6,PtB35,PtB36 and PtB49 could grow regularly on nitrogen-free medium 。PtB6 and PtB49 have the activity of norganic phosphate solubilizing,but this study did not screen out strains with organic phosphorus solubilization ability。Except PtB6,other five strains have different levels of ability to produce IAA,and PtB35 could produce maximum IAA with the concentration of 14。05±1。72μg/mL in it’s  fermentation liquor。Six strains from wheat growth-promoting screening experiments were strains exist different intensity of growth-promoting activity。PtB49 owns all these three abilities of nitrogen fixation,phosphate solubilizing and IAA producing,and it also showed the highest level of growth-promoting among these six strains。
