摘 要:GPX3蛋白与人体抗衰老、糖尿病、克山病、动脉粥样硬化、恶性肿瘤等多种疾病的发生、发展密切作用,作为细胞内重要的抗氧化酶正日益受到医学界的重视[1-4]。为探讨GPX3蛋白的生物学活性及结构与功能的联系。选取酿酒酵母GPX3蛋白为例,从酵母基因数据库中获得了其蛋白基因序列,采用生物信息学方法,对其进行基因序列特征分析,并对其编码的蛋白质从序列特征、理化性质、跨膜结构域、亚细胞定位、高级结构及功能域等方面进行了预测和分析。结果表明,酿酒酵母GPX3蛋白由163个氨基酸组成,在C端存在1个GPX3 保守结构域,其功能与调节细胞生长、抑制凋亡、调节基因转录等功能有关。该蛋白质二级结构中以a一螺旋(31.29)、扩展束(22.09)和无规则卷曲(38.65)为主,不存在任何严谨的跨膜结构,N端不存在信号肽序列,亚细胞定位于细胞质中;该GPX3蛋白与芽殖酵母、德尔布有孢圆酵母的GPX3蛋白相似性较高,进化距离较近。关键词:酿酒酵母;GPX3蛋白基因;功能序列;生物信息学分析9625
Function Sequence of GPX3 Protein Gene
Abstract: In order to study the related functions between the GPX3 protein with human anti-aging, diabetes, Keshan disease, atherosclerosis, cancer and other diseases, choose the GPX3 protein of Saccharomyces cerevisiae as an example.its protein gene sequences obtained from yeast gene database, and Some characters of the GPX3 protein were analyzed and predicted by bioinformatics method,including the composition of amino acid sequence,hydrophobicity,secondary structure and function of protein etc.The results showed that the GPX3 protein of Saccharomyces cerevisiae was composed of 163 amino acids,and contained an GPX3 domain in C end of protein sequence, which functions relevant with regulate cell growth, apoptosis and gene transcription. The secondary structure of the GPX3 protein mainly included a helix (31.29), extended beam (22.09) and random coil (38.65).There is no rigorous transmembrane structure,does not exist N terminal signal peptide sequence, subcellular localization in cytoplasm.The GPX3 protein had higher sequence indentities and closer evolution distance with that from other Saccharomyces cerevisiae including Saccharomyces castellii and Torulaspora delbrueckii.
