摘要:本试验采用国际通用的局部封闭敷贴法进行皮肤致敏试验的研究,初步确定受试物TA(保密药品代号)的致敏性。在试验前24小时,剃去试验动物受试部位的被毛。采用局部封闭敷贴法,先用4只豚鼠确定染毒浓度。正式试验分3组,试验组20只鼠(雌雄各半),诱导浓度为原液,激发浓度为50%稀释液。阴性对照组10个豚鼠(雌雄各半),诱导浓度为丙酮,激发浓度为50%稀释液。阳性对照组(2,4-二硝基氯苯)10个豚鼠(雌雄各半),诱导浓度为8 mg/ml,激发浓度为4mg/ml。各组分别于第0 d、7 d、14 d诱导接触,第28 d激发接触。激发后24h、48h观察豚鼠皮肤过敏反应。阴性对照组和试验组未见红斑、水肿等其他皮肤反应,致敏率均为0%;阳性对照组的动物均可见红斑,其致敏率为100%。32045

Skin Sensitization Test Research
Abstract: Approximately 24 hours before the test,fur was removed by close clipping the test area of the truck of the animals. Buehler test, the appropriate exposure concentration was determined group had twenty albino guinea-pigs(ten male and ten female),the concentration of induction exposure was original liquid,and the concentration of challenge exposure was 50% diluent. The negative control group had ten albino guinea-pigs(five male and five female test animals),the concentration of induction exposure was acetone,and the concentration of challenge exposure 50% diluent. The positive control group (2,4-Dinitrochlorobenzene)had ten albino guinea-pigs(five male and five female),the concentration of induction exposure was 8mg/ml,and the concentration of challenge exposure was 4mg/ml. Induction exposure on day0,day7,day14,challenge exposure on day 28.The albino guinea-pig skin sensitization reaction was observed at 24h,48h skin after challenge. There were no erythema, oedema and other skin changes observed in the negative control group and treatment group after three induction and one challenge exposures,and the
sensitization rate was 0%.Nevertheless,erythema was observed in the animal of the positive control group, and the sensitization rate was 100%.
