摘要:目的:本文旨在研究曲克芦丁对肺腺癌A549细胞的抑制作用并初步探讨其分子机制。方法:A549细胞复苏与传代;划痕实验,研究细胞在不同溶度的曲克芦丁中的迁移情况;收集曲克芦丁处理后的细胞培养液上清检测清除自由基水平;流式细胞仪检测细胞周期实验。
结果:在倒置显微镜下观察曲克芦丁处理过的细胞,发现增殖速度受到明显抑制;划痕实验中A549细胞经曲克芦丁处理后,24h,48h后增殖、迁移速度随化合物浓度增加而降低;在细胞培养液上清清除自由基水平实验中,酶标仪检测出不同浓度曲克芦丁培养液上清液吸光度,来计算出清除率,得出浓度升高清除率升高;流式细胞仪检测细胞周期结果显示,200 uM的曲克芦丁可以将细胞周期阻滞在G1期。51847
结论:曲克芦丁能够通过靶向A549细胞的G1期,抑制肿瘤细胞的增殖,这种作用可能与其清除自由基和抗氧化活性相关。

Inhibitory role of troxerutin on human lung cancer A549 cells
Objective: This paper aims to investigate the inhibitory effect of troxerutin on human lung cancer A549 cell and to make clear the possible mechanisms.
Methods: Thaw and passage the A549 cells; Wound scratch test was performed to test cell migration after troxerutin treatment with different concentration; Supernatant in different groups were collected to detect anti-oxidation activity; Flow cytometry was performed to identify cell cycle.
Results: Under inverted microscope, we found troxerutin significantly inhibited proliferation of A549 cells; The migration of A549 was suppressed by troxerutin in dose-dependent manner as shown in wound scratch test; The result of anti-oxidation experiments showed that troxerutin could eliminate free radicals dramatically in comparison with normal A549 cells; Meanwhile, the analysis of flow cytometry suggested that cell cycle of A549 cells was blocked in G1 phase after administration of 200 uM troxerutin .
Conclusion:Troxerutin can inhibit the proliferation of A549 cells by targeting G1 phase, and the activity of anti-oxidation might be involved in this process.
