摘要:干扰素(Interferon, IFN)是一类多功能的细胞因子,在抵抗病毒感染、抑制细胞增殖和免疫调控方面扮演着重要角色。干扰素调控因子家族作为调节干扰素表达的转录因子被发现,随着其在先天性免疫和获得性免疫中的关键作用而得到越来越多的关注。干扰素调控因子5(IRF5)是家族成员之一,也是免疫系统中一个重要的转录因子,在抗病毒反应中表现出多种生物学功能。本实验以长白猪、大白猪、松辽黑猪三个品种为研究对象,初步分析了猪IRF5基因的分子特征,并针对IRF5基因的外显子区域,设计特异性引物,运用PCR扩增和PCR产物直接测序技术及DNAMAN、CHROMAS等生物学软件,在IRF5基因外显子7区域筛选出一个SNP位点(g.10764 T>C),这为进一步将IRF5基因作为与猪免疫性状相关的研究提供了科学参考依据。28368 毕业论文关键词:猪;IRF5;单核苷酸多态性
Identification analysis of porcine interferon regulatory factor 5 (IRF5) gene
Abstract: Interferons (IFNs) are a family of multifunctional cytokines that potently demonstrate antiviral, ant proliferative and immunomodulatory activities. Interferon regulatory factors (IRFs) were identified originally as transcription factors in the regulation of interferon expression and has been received more and more attention for its key role that links innate and adaptive immune response. Interferon regulatory factor 5 (IRF5) gene is one of IRFs family and is also a crucial transcription factor in immune system, it has shown its functionally perse roles in the regulation of antiviral responses. In this experiment, three breeds of pig which include Landrace, Large white and Songliao black pig were selected to analyze the molecular characterization of IRF5 gene firstly. In addition, we designed the specific primer pairs fort exons region of porcine IRF5 gene and identified a single nucleotide polymorphism in exon 7 (g.10764 T>C) by PCR amplification, directly sequencing technology for PCR products and DNAMAN, CHROMAS software. These results should provide scientific reference for the further association study between IRF5 gene and immune traits in pig.
