摘要:【目的】对水稻叶色斑马叶突变体进行遗传分析与基因定位,可以为阐明水稻质体发育和叶绿素合成的分子机制奠定基础。【方法】本研究将水稻斑马叶突变体zebra leaf 3与武运梗7号和02428分别杂交,构建F2群体,进行遗传连锁分析。斑马叶突变体zl3材料是籼稻9311经过甲基磺酸乙酯(EMS)诱变所得。剪取3叶期后5和8天的叶片,观察表型。【结果】测定结果表明,zl3基因单碱基的突变导致了苗期淡黄条纹叶的产生,同时伴随着叶绿体发育的异常,叶绿素含量和胡萝卜素含量相比野生型减少。遗传分析之后的结论是此突变性状是由一对隐性核基因控制的。利用zl3/02428 F2群体,可以将该基因定位于第8染色体短臂上标记VF3和VF4之间,区间大小约为58.2kb范围内。【结论】水稻斑马叶突变体zl3突变性状受1对隐性核基因控制,基因定位于第8染色体上58.2kb区间内。zl3的发现旨在为克隆该基因并对研究叶绿体发育过程提供参考。29143 毕业论文关键词: 水稻;叶色突变体;图位克隆;叶绿体
Fine mapping of zebra leaf color mutant gene zebra leaf 3 in rice
Abstract: [Objectives] The aim is to identify rice pale zebra leaf mutant zebra leaf 3(zl3), localize the target gene and lay the function for the study of the molecular of chlorophyll biosynthesis and plastid development. [Method] The rice zebra leaf mutant zl3 crossed with 02428 respectively to construct F2 population, conduct genetic linkage analysis. Zebra leaf mutant zl3 was isolated from an EMS-induced indica rice 9311 mutant bank. The phenotypes of the leaves that are 5 and 8 days respectively after 3-leaf stage were observed. [Results] 3-leaf stage photosynthetic pigment content determination results showed that chlorophyll a, chlorophyll b and carotenoid content were significantly lower than the wild type. By genetic analysis concluded that the zebra leaf phenotype was controlled by a single recessive gene. By using zl3/02428 F2 population, the target gene can be located on chromosome 8 and positioned between markers VF3 and VF4, and the physical distance was about 58.2kb. [Conclusions] The phenotype of zl3 was controlled by a single recessive gene. zl3 was found on chromosome 8 within the physical distance was about 58.2kb.The mapping of zl3 provides references for the cloning of the gene and the functional study of chlorophyll synthesis and chloroplast development.
