摘要:本文研究了热处理对奶牛卵巢颗粒细胞HO-1基因表达的影响。首先采集健康奶牛卵巢组织,并对卵巢中的颗粒细胞进行体外培养,然后采用40°C热处理细胞,从而测定热处理对卵巢颗粒细胞HO-1基因mRNA及蛋白水平的表达情况,并进一步测定细胞中HO-1代谢产物一氧化碳(CO)及细胞内信使环磷酸鸟苷(cGMP)水平,旨在揭示HO-1在热应激状态下的表达情况为进一步研究应激状态下卵泡微环境提供一定的参考依据。29476 毕业论文关键词:奶牛;热处理;卵巢颗粒细胞;HO-1;细胞保护
Effect of heat stress on expression of HO-1in ovarian granulosa cells in vitro cultured
Abstract: In this study, the effect of heat treatment on HO-1 gene expression in granulosa cell was detected. The dairy cow granulosa cells(GCs) were cultured by using tissue inoculation method. Then the dairy cow granulosa cells were treated by heat. After heat treatment, the content of CO and cGMP in granulosa cell were determined. The objective of this study was to reveal the protective mechanism of HO-1, and to provide some references for identifying the micro-environment of follicle under stress condition.
