摘要:黄曲霉是食物的腐败变质的主要原因之一,控制食品腐败造成的食品安全问题是十分必要的。紫苏醛是从紫苏中提取的挥发油中的一种,通过紫苏醛对黄曲霉细胞周期及细胞凋亡的影响,从而来探明紫苏醛抗黄曲霉的作用机理,对于进一步开发天然植物防腐剂有重要的作用。本实验主要通过紫苏醛对黄曲霉细胞膜麦角甾醇含量的测定来确定紫苏醛是否对黄曲霉有作用,从而进一步研究不同浓度紫苏醛对黄曲霉细胞周期及细胞凋亡的影响。结果显示,麦角甾醇实验中,最终得到的数据显示麦角甾醇的含量随着紫苏醛浓度的升高而降低。在紫苏醛对黄曲霉细胞周期的实验中,随时间的增加,可以看出细胞周期越来越集中于G1期,这显示紫苏醛能够影响黄曲霉的细胞分裂;在细胞凋亡的实验中可以看出紫苏醛能够引起黄曲霉的早期凋亡。这一系列实验对于开发新的天然植物防腐剂有重要的理论指导意义。34669 毕业论文关键词:紫苏醛;黄曲霉;细胞周期;细胞凋亡
The Influence of Perillaldehyde on Cell Cycle and Cell Apoptosis of Aspergillus Flavus
Abstract: A. flavus is one of the main reasons for the deterioration of food, control the food corruption caused by food safety problem is very necessary. Perillaldehyde, which is one of the essential oil extracted from perilla by perillaldehyde influence on aflatoxin cell cycle and cell apoptosis, and to explore the effect of perillaldehyde resistance to A. flavus mechanism, plays a key role for the further development of natural antiseptic. This experiment mainly through the ergosterol content determination of perillaldehyde to A. flavus cell membrane has the function to determine whether the perillaldehyde to A. flavus, thus further study on different concentrations of perillaldehyde on aflatoxin cell cycle and cell apoptosis. Results indicated that ergosterol experiment, the resulting data show the ergosterol content and decreased with the increase of concentration of perillaldehyde. In perillaldehyde to A. flavus cell cycle experiment, with the increase of the time, it can be seen that the cell cycle is more and more focused on G1 phase, this shows that perillaldehyde can affect the A. flavus cell pision; it also can be seen that in the apoptosis of experiments perillaldehyde can cause early apoptosis of A. flavus. This series of experiments for the development of new natural preservatives have important theoretical guiding significance.

紫苏[Perilla frutescens (Linn.) Britt]是一种具有重要的生理功能和保健价值的植物,既可药用也可当蔬菜食用,在我国有悠久的历史,属唇形科一年生草本植物,又名苏子、回回苏、红紫苏、红苏、赤苏等,世界各地均有种植[1]。紫苏籽油中含有人体所需八种氨基酸,同时还含有婴幼儿所需九种必须氨基酸,含有蛋白质、β-胡萝卜素、VB1、VB2、VC、PP、Ca、Zn、Fe、Cu、Mo、Mn等多种活性元素及矿物质[2,3,4]。紫苏中含有多种黄酮类、甙类、萜类等化合物,具有调节血脂、降血压、抗氧化和抗癌等作用。Gisho研究发现,紫苏精油中的紫苏醛有抑制细菌生长的作用[5];Kang等通过对紫苏精油抗菌活性进行研究,发现其中紫苏醛对多种细胞有抑制作用,能有效防止由沙门氏菌属引起的海产品食物中毒[6];Koga等认为紫苏醛与其他丁香油酚可以有效的抑制绿脓杆菌的传染[7]。
