摘 要:以安琪酵母为材料,用酵母完全培养基(YPD培养基)进行培养。通过稀释涂布平板法挑选出纯菌落。经过镜检之后,选取活性高的菌株进行扩大培养。对培养至对数期的酵母,用不同浓度的CdCl2,ZnCl2 对酿酒酵母进行48 h胁迫处理,对刺激处理后酵母进行活细胞/死细胞计数比,观察生长曲线,并破碎酵母细胞壁,提取酵母基因组DNA,对处理过的酵母DNA进行凝胶电泳实验。34986 毕业论文关键字:酿酒酵母;CdCl2; ZnCl2; 胁迫处理;基因组
Different Concentration of CdCl2,ZnCl2 Effect on Saccharomyces Cerevisiae Genome
Abstract: The experiment used Angel yeast as the material, Yeast was incubated by yeast culture medium (YPD medium). Through the method of dilution coated tablet single yeast colony was selected. After a detection, the yeast strain were cultured in YPD medium. The yeasts were cultured to the logarithmic phase of yeast, and were stressed with different concentration of CdCl2, ZnCl2 for 48 h. The stressed yeast cells were counted, and yeast cells wall were broken, yeast were extracted genomic DNA and analyzed using DNA gelectrophoresis technique.
