摘要:随着我国经济的快速发展和人们生活水平的逐渐提高,染料废水的排放量也随之增多,因而造成的环境污染也变得日趋严重。染料废水的处理方法有多种,主要有生物处理法,化学处理法,物理处理法等,但由于生物处理法的优点较多而被广泛应用于日常生产和生活中。本研究采集了不同地区的土壤,通过一定梯度的稀释后,利用含有亚碲酸钾的培养基筛选出能够使菌落变黑的一些菌株,从而获取具有电子传递功能的菌株用于染料降解。通过对培养基使用四区划线法来纯化获得单一的菌种,本研究共得到了7株变黑菌落的菌株。最后对分离所得到的染料废水降解菌菌株进行了初步的功能鉴定和形态学观察,并对其中降解效果较好的一株菌,利用现代分子生物学技术进行扩增菌体的16S rRNA基因,从而确定菌种的种属地位,结果发现其为Escherichia hermannii.37243 毕业论文关键词:染料废水;降解菌;废水处理
Dye wastewater degradation bacteria isolation and molecular identification
Abstract: With the increasing of wastewater drainage in China because of the rapid development of economic and increasing of the standard of living,the water pollution becomes more and more serious. There are different kinds of dyes wastewater treatment mainly include biological treatment, chemical treatment, physical treatment, etc., but due to the advantages of biological treatment method is more widely used in daily life and production. This study collected in different parts of the soil, through a certain gradient dilution, using culture medium containing potassium tellurite can make colonies black screened out some strains, thus obtain strains with electron transfer functions used in dye degradation .Through the use of the four pisions line method purifies the single species, this study, a total of 7 of black colonies of strain Finally, the separation of the function of the dye wastewater degradation bacteria strains has carried on the preliminary appraisal and morphology observation, and the degradation of the bacterial strains with better effect, use of modern molecular biology techniques for amplification of bacteria 16S rRNA genes, and thus determine the species of genus status, found that it is Escherichia hermannii.
