摘要:利用响应面分析法优化莲蓬壳总黄酮提取工艺条件。在单因素试验基础上,以提取时间,提取温度,料液比及乙醇浓度为响应因子,总黄酮提取得率为响应值,进行四因素三水平响应面法分析,应用Design-Expert 8.0.6软件,模拟得到二次多项式回归方程的预测模型,并确定乙醇提取莲蓬壳总黄酮的最佳工艺条件为∶液料比40 g/ml,乙醇浓度为48%,提取温度60 ℃和提取时间10 min。在最佳工艺条件下,每提取1次,莲蓬壳总黄酮提取量为8.322 g/100 g,接近于预测值8.642 g/100 g,说明采用响应面法优化得到的提取条件可靠。54357

Abstract: Response Surface Analysis (RSA) was applied to optimize the extraction of the total flavonoids of Lotus receptacle. On the basis of single-factor test, with extraction time, extraction temperature, solvent/material ratio and ethanol concentration for the response factor, total flavonoids obtained rate in response to a value, analyses the response surface methodology (RSM) with 4 factors and 3 levels. Design-Expert 8.0.6 software was used to fit experimental data and a quadratic polynomial regression model was developed for the yield of the total flavonoids of lotus receptacle as a response to the parameters. Results showed that the optimal value of extraction conditions were as follows:solvent /material ratio 40 g/ml, ethanol concentration 48%, extraction temperature 60 °C and extraction time 10 min. Under such conditions and extraction 1 time, the yield of the total flavonoids of lotus receptacle was up to 8.322 g/100 g, which is close to the predictive yield (8.642 g/100 g), suggesting that the optimal extraction by conditions the response surface methodology( RSM) are reliable.
