摘要本次研究使用80个湖羊个体为实验样本,通过PCR-SSCP技术寻找湖羊的PPP1CB基因是否存在多态位点,另外,运用DNA测序技术来检测PPP1CB基因的多态位点的位置以及其碱基变换情况。在找到PPP1CB基因的单核苷酸多态位点的基础上,运用生物信息学技术,探索PPP1CB基因的多态性与湖羊的体高、体长、胸围、管围等体尺性状是否存在相关性,并且总结出PPP1CB基因的多态性对湖羊生长发育状况的影响程度,为湖羊的繁殖育种、产肉性能的提升提供科学的研究基础。经过实验研究发现,湖羊的PPP1CB基因在P5位点存在着1个单核苷酸多态位点,并且是低度多态,另外A5A5是占优势的基因型。经过分析研究知道,PPP1CB基因的多态性对湖羊的体尺性状以及生长发育产生了一定的影响。因此,在对湖羊进行选育的过程中,可以将PPP1CB基因的突变位点作为一种湖羊育种的参考依据。71687

Preliminary Analysis of Genetic Diversity of PPP1CB Gene in Hu Sheep
Abstract In this study we use 80 Hu Sheep inpiduals as the experimental sample. We look for the PPP1CB gene in Hu Sheep Whether there are polymorphic loci by the technology of PCR-SSCP. In addition, We detected the location of the polymorphic of the PPP1CB gene and its base transformation with DNA sequencing technology. Based on the single nucleotide polymorphism of the PPP1CB gene by bioinformatics technology to explore weather it has the relationship between the polymorphismof PPP1CB gene with Hu Sheep’s body height, body length, chest circumference, round tube and other aspects of body condition. Then summed up the influence degree of polymorphism of PPP1CB gene on growth and development of Hu Sheep to provide scientific research foundation for the breeding of Hu Sheep and meat production performance. After the experimental study found There are one single nucleotide polymorphisms of PPP1CB gene of Hu Sheep and it’s low polymorphism. In addition, A5A5 is the dominant genotype. Through analysis and research to know that the polymorphism of PPP1CB gene has a certain impact on body size traits and the growth in Hu sheep.Therefore, we can use the transformation of polymorphic sites of PPP1CB gene as a selection basis in the process of breeding of Hu Sheep.
