    摘要:本课题以青钱柳原叶为材料,采用热浸法对青钱柳有效成分多糖进行提取。通过单因素实验研究了,提取时间、提取次数、提取温度及料液比对青钱柳多糖得率的影响。并结合正交试验确定了热浸提法提取青钱柳多糖的最佳工艺参数,即提取温度为100℃,提取次数为2次,提取时间为200min,液料比为1:16。在上述提取条件下,热水浸提法提取青钱柳多糖得率可达到5.92%。然后根据上述最佳工艺提取青钱柳多糖,用其对实验性高血糖小鼠进行灌胃,观察小鼠的血糖的变化,以确定青钱柳多糖降血糖活性。结果表明青钱柳多糖高、中、低剂量组与对照组血糖值有著性差异,由此说明青钱柳多糖可以显著提高小鼠对葡萄糖的糖耐量。并通过青钱柳多糖高、中、低剂量组的降糖效果的对比,初步确定当多糖用量为4g/kg时降糖效果显著,多糖利用率较高。同时采用体积表面积直接换算法计算人体青钱柳多糖最佳用量为19.59g~24.84g。在青钱柳多糖应用方面,采用感官评定和正交试验法对青钱柳多糖功能性饮料的配方进行了研究,确定青钱柳多糖功能性饮料的最佳配方,即青钱柳多糖用量5%、柠檬酸用量0.014%、蔗糖用量3.5%。该产品为淡褐色、澄清饮料,pH为4.6,适用于高血糖、高血脂症及普通人群食用。7803

    Study of extraction of active components in Cyclocarya paliurus and its application in soft drinks

    Abstract :In this study, polysaccharides were extracted from cyclocarya paliurus(CP) leaves by hot-dip method. The impacts of extraction time, extraction times, extraction tempe- rature and solid-liquid ratio on recovery ratio of CP polysaccharides were determined by single factor experiment. And then orthogonal test was used to determine the optimum process parameters for extraction of polysaccharides by hot-dip method. The result showed that optimum extraction temperature was 100℃,2 times, time 200min,solid-liquid ratio 1:16. In the extraction conditions above, recovery ratio of CP polysaccharides was up to 5.92% by the hot water extraction. The guinea pigs with high blood sugar were fed with the CP polysaccharides by the method of gavages administration. Then changes of blood sugar in guinea pigs were observed to determine CP polysaccharides’ hypoglycemic activity. The results showed that the blood glucose of high, medium and low-dose group of CP polysaccharides were significantly different from model group. And this meant that CP polysaccharides can significantly improve glucose tolerance in guinea pigs. And the comparison of hypoglycemic activity in high, medium and low-dose group showed that hypoglycemic activity was the best when the dosage of polysaccharides is 0.4g/kg. In this condition, the utility ratio of CP polysaccharides was also relatively high. And then method of mass- surface conversion was used to calculate the optimum dosage of CP polysaccharides applied in human bodies. The result showed that the optimum dosage of CP polysaccharides applied in human bodies was 19.59g~24.84g. In terms of CP polysaccharides’application, sensory evaluation and orthogonal test were used to determine the recipe of functional beverage prepared by CP polysaccharides. The optimum recipe was: CP polysaccharides 5%, citric acid 0.014%, sucrose 3.5%. The beverage was light brown, clear and its pH was 4.6. It was suitable for hyperglycemia, hyperlipidemia and ordinary people to drink.
