目的： 探讨SIRT2过表达对人肺癌细胞迁移和侵袭力的影响。 方法：SIRT2表达载体的转染和western blot鉴定，Transwell实验评估迁移力和侵袭力；采集临床肺癌组织样本，分原发灶和转移灶两组，免疫组织化学法检测各样本中SIRT2的表达，统计分析Sirt2表达对肺癌转移的影响。结果：成功的构建SIRT2过表达人肺细胞株，经Western blot 鉴定转染成功后，通过Transwell实验可知，SIRT2过表达组比对照组相比细胞数量明显增多，SIRT2过表达促进人肺癌细胞迁移和侵袭力。免疫组化实验可知，转移率有显著性差异（P<0。05），转移组的SIRT2表达量高于非转移组  结论 ：SIRT2过表达能促进人肺癌细胞迁移和侵袭力。89707

Objective： reacher to study the effects of SIRT2 overexpression on migration and invasion of human lung cancer cells 。  Methods: plasmid transfection and Western blot confirmed the expression of SIRT2, Transwll migration and invasion experiment evaluation; collection of clinical lung cancer tissue samples, pided into primary and metastatic foci of two groups, the expression of SIRT2 was detected by immunohistochemistry in all samples, statistical analysis of the expression of Sirt2 on lung cancer metastasis effect。   Results: the construction of over expression of SIRT2 in human lung cell line, blot was identified by Western after the success of Transwell, through the experiment, the overexpression of SIRT2 group than in the control group compared to the number of cells increased significantly, immunohistochemical experiment showed that, There was significant difference in mobility(p<0。05),SIRT2 in metastatic group was higher than that in non metastasis group。 Conclusion: overexpression of SIRT2 can promote the migration and invasion of human lung cancer cells。
