摘    要

目的 分析三磷酸腺苷结合盒转运子A1(ATP Binding Cassette Subfamily A Member 1, ABCA1)基因的rs2000069、rs4743764、rs4149336位点与汉族人群肥胖相关性。

方法 采用整群随机抽样的方法抽取年龄≥40岁的汉族成年人为研究对象。由受过专业训练的医生和护士采集体检信息,问卷调查收集调查人群的生活行为方式等信息。采用聚合酶链式反应-连接酶检测反应(polymerase chain reaction-ligase detection reaction,PCR-LDR)技术对ABCA1基因的rs2000069、rs4743764、rs4149336位点分型检测。采用χ2检验分析各位点基因型在BMI正常、超重和肥胖三组中的分布差异;采用多重线性回归模型分析ABCA1基因多态性对BMI和血脂水平的影响。85438

结果 本研究共收集到符合要求的样本2203人,其中男性1003例,女性1200例,男性平均年龄为60。08±11。58岁,女性的平均年龄为58。22±11。03岁。采用PCR-LDR技术进行基因位点分型检测。卡方检验结果显示rs2000069、rs4743764、rs4149336位点基因型在BMI正常、超重、肥胖组的分布有显著差异。多重线性回归显示,在调整了年龄、性别和腰围后rs2000069、rs4743764 、rs4149336这3个位点多态性均与BMI显著相关;rs4149336位点多态性与TC、LDL-C显著正相关,而rs2000069、rs4743764、rs4149336位点均与TG、HDL-C无关。源Q于W优H尔J论K文M网WwW.youeRw.com 原文+QQ75201.,8766

结论  ABCA1基因rs2000069、rs4743764、rs4149336位点均与BMI相关,而rs4149336位点与TC、LDL-C显著正相关,这三个位点可能与超重/肥胖发生风险有关。

Abstract

Object:to investigate the association between the gene polymorphisms of rs2000069、rs4743764、rs4149336 in ABCA1 (ATP Binding Cassette Subfamily A Member 1, ABCA1) gene and obesity in Chinese Han population。 

Methods: The Han adults aged ≥40 years old were selected as the research subjects by means of random sampling。 By the professional training of doctors and nurses to collect medical information, questionnaire survey to collect information on the behavior of people surveyed。 The rs200069, rs4743764 and rs4149336 loci of ABCA1 gene were detected by polymerase chain reaction-ligase detection (PCR-LDR) technique。 The distribution of ABCA1 gene polymorphism on BMI and blood lipid levels was analyzed by multiple linear regression model in the normal, overweight and obese groups of BMI。

Results: In this study, a total of 2203 samples were collected, including 1003 males and 1200 females, with an average age of 60。08 ± 11。58 males and an average age of 58。22 ± 11。03 for females。 PCR-LDR technique was used to detect the loci, and the primers, probe sequences and product length of each locus were obtained。 Multiple linear regression showed that the polymorphisms of rs2000069, rs4743764 and rs4149336 were significantly correlated with BMI after adjustment of age, sex and waist circumference。 Rs4149336 polymorphism was positively correlated with TC and LDL-C, while rs2000069, rs4743764, rs4149336 Points are independent of TG, HDL-C。

Conclusions: The ABCA1 gene rs2000069, rs4743764 and rs4149336 were all associated with BMI, while the rs4149336 locus was positively correlated with TC and LDL-C, which may be related to the risk of overweight / obesity。
