摘要:本试验旨在进一步探索长期灌胃添加低剂量纳米ZnO对小鼠发育及免疫功能的作用。试验分为三组:1)对照组,灌胃生理盐水;2)灌胃每千克体重25mg纳米ZnO(以锌计);3)灌胃每千克体重100mg纳米ZnO(以锌计),每组三个重复,每个重复8只21天断奶仔鼠。13周结束,分别从生长、免疫等方面系统评价长期添加低剂量纳米ZnO的安全性。结果表明:长期灌胃没千克体重25mg及100mg纳米ZnO对小鼠的生长无显著影响 (P>0.05);可显著增加十二指肠绒毛的高度及隐窝深度(P<0.05)以优化肠形态。在元素沉积方面,灌胃添加每千克体重25mg及100mg 纳米ZnO均可显著增加肝脏中Zn的含量及肾脏中Fe的含量(P <0.05),对肌肉中元素沉积均无显著影响。此外,灌胃100mg每千克体重纳米ZnO显著增加肾脏中Fe的含量并且使血液内淋巴细胞数显著增多(P <0.05)。本研究结果提示,长期灌胃添加每千克体重25mg和100 mg 纳米ZnO对小鼠不仅无明显毒性作用,而且在肠形态及免疫方面存在可能的促进作用。29798

Effects of Nano Zinc Oxide on Growth Performance and Immunity in Mice via Long-Term Intragastric Administration
Abstract:This study aimed at further investigtating effects of low dose nano Zinc Oxide (nano-ZnO) on the growth and immune function of mice via long-term intragastric administration for 13 weeks. The experiment was pided into three groups randomly, which were 1)the control group, treated with physiological saline; 2)25mg/kg nano-ZnO group: intragastric administration with 25 mg zinc as the form of nano zinc oxide /kg body weight; 3) 100mg/kg nano-ZnO group: intragastric administration with 100mg zinc as the form of nano-ZnO /kg body weight. Each group contained 3 repeats, eight mice for each repeat. All mice in the experiment weaned at 21 days. After 13 weeks of experiment, the safety of long-term low dose nano-ZnO was evaluated from the aspects of growth, immunity and so on. The results showed that the gastric 25 and100 mg/kg body weight of nano-ZnO had no significant effect on the growth of mice (P>0.05); optimized intestinal morphology by enhancing the height of duodenal villus and the crypt depth significantly (P <0.05).In terms of element deposition the gastric 25 and 100mg/kg body weight of nano-ZnO significantly increased the content of Zn in liver and the content of Fe in kidney and showed no effect on element deposition in muscle. In addition, the gastric 100mg/kg body weight of nano-ZnO significantly increased the content of Fe in kidney and the number of lymphocytes in the blood (P < 0.05).Our results demonstrated that the long-term gastric 25 and 100mg/kg body weight of nano-ZnO has no obvious toxic effect, but it can probably promote intestinal morphology and immunity.
