摘要由于年龄增长及高脂高糖的饮食习惯,糖尿病已经成为当今人类健康的重大疾病之一。研究表明,大量糖尿病患者的致死因素是心血管功能的紊乱,而作为最先接触血液的屏障,位于血管内壁的内皮细胞功能是关系心血管功能的重要指标。内皮层的老化可打乱内皮功能,引发一系列心血管疾病。大量文献表明,槲皮素可以通过增强毛细血管抵抗力、扩张冠状动脉、增强动脉血流量等,对冠心病和高血压等疾病有辅助治疗的作用。但是,关于槲皮素对糖尿病中内皮细胞老化影响的研究很少。基于此,我们拟利用人脐静脉血管内皮细胞为培养模型,研究高糖处理对血管内皮老化的影响以及槲皮素的干预作用并探讨其分子机制,为临床治疗高糖症及血管老化相关疾病提供理论依据。51003

Quercetin on vascular endothelial cells
Abstract
Diabetes has become one of the major diseases of human health, because of aging and high-fat or high-glucose diet. Studies have shown that cardiovascular disorders are major cause of mortality in diabetes mellitus. As the first barrier to inflammation in blood, vascular to maintain endothelial cells, which lie inner lining of blood vessels, are vital indicator to maintain endothelial homeostasis and functions. Senescence of endothelial may disrupt endothelial function, and trigger a series of cardiovascular disease. A lot of literature suggests that quercetin can enhance capillary resistance, expansion of coronary artery, enhance blood flow and so on. It has been used for therapy of coronary heart disease and hypertension. However, litter is known about the role of quercetin on endothelial senescence in diabetes mellitus. Based on this, we adopted human umbilical vein endothelial cells (HUVECs) as the in vitro model, used high glucose treatment to mimic diabetes mellitus, to investgate effect of high glucose on endothelial senescence and the interferential impact of quercetin ,and to explore the underlying molecular mechanism. This study will provide new clues for clinical treatment of hyperglycemia and endothelial senescence-related cardiovascular diseases.
