摘要:目的:本文旨在检测PSPC对肺腺癌A549迁移和细胞周期的影响,以及其抗氧化活性, 以此来研究PSPC抑制细胞生长的机制,对研发新的抗癌药,临床治疗肺癌都有重要意义。
方法:倒置显微镜下观察细胞形态;通过设计划痕试验,观察细胞在含不同浓度PSPC的培养液中迁移情况;收集不同处理组的细胞培养液上清,检测其抗氧化活性水平;流式细胞术检测细胞周期分布状况。

结果:形态观察显示,PSPC处理过的A549细胞,增殖速度受到一定的抑制;在划痕实验中,A549细胞经过PSPC处理24h、48h后,细胞生长迁移的速度随化合物浓度的升高而降低;自由基清除实验结果显示,与正常组的细胞相比,PSPC处理后的培养液上清中,自由基清除能力显著升高, 并且PSPC清除自由基的活性具有浓度和时间依赖性;通过流式细胞仪检测细胞周期, 我们发现100ug/L的PSPC处理A549细胞后,显著提高G1期的细胞比例,而200ug/L 的PSPC使S期细胞增加,两种浓度均使G2/M期细胞比例降低。
结论:紫甘薯花青素(PSPC)能够抑制细胞的增殖,有较强清除A549细胞自由基的能力,且对A549细胞周期有着显著抑制的作用。

Inhibitory effect of purple sweet potato color on the growth of human cancer cells
Abstract Objective: This paper aims to study the inhibiting effect of purple sweet potato color (PSPC) on human lung cancer cell line A549 cells, and the anti-oxidative ability, to clarify the underlying mechanisms, which are important for the development of new anti-cancer drugs, and also supply theory support for the clinical therapy for human lung cancer.
Methods: Cell morphology was observed under a inverted microscope; the effects of different concentrations of drugs on cell migration were determined through the scratch test; Supernatant were collected to test the anti-oxidative activity of PSPC; Cellular cycle distribution was analyzed by flow cytometry.
Content: to master the culture of lung adenocarcinoma A549 cells; To detect the inhibitory effect of PSPC on migration and cell cycle; To master the commonly used technology of cell biology by discussing molecular mechanism.
Results: Results of cellular morphology showed that the proliferation of A549 was inhibited by PSPC. In the scratch test, A549 cells were administered with 100 and 200 ug/L PSPC for 24 and 48 hours, and cell migration was inhibited with the increase of concentration of PSPC. Besides, compared with normal group, the free radicals in A549 were significantly cleared by PSPC in both dose and time dependent manner. In addition, the result of cell cycle distribution showed that the number of cells in G1 phase was increased by PSPC at 100ug/L, while the number of cells in S phase was elevated by 200ug/L PSPC, and both two concentration decrease cell number in G2/M phase.
Conclusion: PSPC inhibits cell proliferation and exhibits strong ability of free radical removal in A549 cells, and has a significant obstructive effect on A549 cell cycle.
