摘要本实验研究探寻利用超声辅助提取悬铃木叶中总黄酮的提取工艺的最佳条件及其体外抗氧化活性。实验利用乙醇作为提取溶剂,通过超声辅助提取悬铃木叶中的总黄酮,对超声频率、超声时间、乙醇体积分数以及料液比进行单因素试验,研究各因素对其总黄酮提取量的影响作用,并进一步通过响应曲面法优化悬铃木叶中总黄酮的提取工艺条件,通过统计分析得到最佳提取工艺条件为:超声频率67.7Hz,超声时间50.5min,乙醇体积分数30%,料液比1:53.9,在此工艺条件下悬铃木叶中总黄酮的提取量为8.1278mg/g,与预测值接近,无显著差异,因此响应曲面法优化悬铃木叶中总黄酮的提取工艺条件合理可行,具有一定的实际应用价值。通过对悬铃木叶中总黄酮的提取液进行体外抗氧化性实验,表明其具有较强的抗氧化活性,尤其对清除羟自由基有着较强的作用,且其抗氧化性随着黄酮含量的增加而有着明显的增加,具有进一步研究利用的价值。69844

Optimization of Ultrasonic Assisted Extraction of Total Flavonoids from the Leaves of Platanus acerifolia by Using Response Surface Method
Abstract The study aimed to discuss the optimum technology condition for the ultrasonic assised extraction of total flavonoids from the leaves of Platanus acerifolia and anti-oxidation activity. The total flavonoids in the leaves of Platanus acerifolia were extracted by ultrasonic assisted extraction using ethanol. The ultrasonic frequency, ultrasonic time, ethanol concentration and the ratio of material to extraction reagent were selected as the influencing factors for the single-factor experiments. The response surface methed was used based on the results of single-factor test to study the optimization of ultrasonic assisted extraction of total flavonoids from the leaves of Platanus acerifolia. The optimum technology condition for ultrasonic assisted extraction of total flavonoids from the leaves of Platanus acerifolia was determined as follows: ultrasonic frequency was 67.7Hz, ultrasonic time was 50.5min, ethanol concentration was 30%, the ratio of material to extraction reagent was 1:53.9. Under the optimum technology, the extraction efficiency of total flavonoids reached 8.1278mg/g. The total flavonoids from the leaves of Platanus acerifolia had obvious anti-oxidation property, especially in scavenging effects on hydroxyl free radicals. And the inoxidizability increased in a concentration-dependent fashion.
