摘 要:本实验以北京农科院提供的京-17品系小麦种子为材料,以lacl3母液为处理液,研究了不同浓度lacl3处理液对小麦的根的生长状况和小麦根尖DNA甲基化的影响,以期为其表观遗传机理提供参考。对处理后的小麦根长进行统计,并用graph 5软件作图并分析。结果表明低浓度的lacl3 处理液能促进小麦主根长,而高浓度的则抑制其生长。然后选择与对照组差异明显的浓度处理组,采用CTAB法提取其根尖DNA,再用HAPII酶处理,用PCR技术扩增目的基因并对其进行琼脂糖凝胶电泳。结果表明有些基因条带变暗,说明小麦根尖出现了部分甲基化或者完全甲基化的情况,证明了稀土元素镧促进了小麦根尖DNA的甲基化。34206 毕业论文关键字:小麦;稀土元素镧;DNA甲基化;graph 5软件;PCR检测
Rare earth element La effects on wheat root tip of DNA methylation
Abstract:This experiment provided in Beijing academy of Beijing - 17 strains of wheat seeds as the material, with lacl3 mother liquid to process fluid, different concentration of lacl3 processing was studied in wheat root growth and root tip of wheat, the effects of DNA methylation in order to provide reference for its epigenetic mechanism. To statistics of treated wheat root length, and the graph 5 drawing and analysis software. Results show that the low concentration of lacl3 processing liquid can promote the wheat root length, and high concentration of inhibit its growth. Then select have significant difference, the concentration of the treatment group with control group the CTAB method is adopted to extract the root tip of DNA, reoccupy HAPII enzyme treatment, genes using PCR amplification and agarose gel electrophoresis. Results show that some genes with dark, wheat root tip appeared some methylation or complete methylation, proved that the rare earth element lanthanum promoted the wheat root tip of DNA methylation.
