摘要:细胞程序性死亡对于梨石细胞形成具有重要的生物学意义。Metacaspase蛋白酶是植物细胞中的类Caspase蛋白酶,在植物细胞程序性死亡中起到类似Caspase的关键调控作用。本研究通过梨全基因组数据库,利用生物信息学方法对梨12条Metacaspase基因家族进行了蛋白序列的系统分析和染色体定位分析,以及氨基酸的组成、理化性质和二级结构分析,此外还分析了梨与拟南芥之间的基因家族联系,最后通过半定量RT-PCR进行了基因的表达分析。结果显示12条蛋白序列分成了两个类型,基因组定位在7条染色体上,比拟南芥更为分散。蛋白质二级结构分析发现氨基酸序列以无规则卷曲为主要组成部分。基因表达显示在果肉和根中的表达量最高。本文的研究结果对梨Metacaspase基因家族的进一步功能分析提供了重要基础。38824 毕业论文关键词:梨;Metacaspase基因家族;生物信息学
Bioinformatics analysis of the Metacaspase gene family in Pear
Abstract:The programmed cell death has a very important biological significance for the formation of pear stone cells. Metacaspase protease is the Caspase-like protease in plant cells, and it plays a key role in programmed cell death which similar to Caspase-like protease. This study is conducted by pear whole genome database, and the bioinformatics method is used to analyze the protein sequence, chromosome localization, amino acid composition, protein secondary structure and the gene family relationship of 12 sequences of Metacaspase gene family. In the end, the study perform the RT-PCR experiment to analyze the gene expression. The results shows that 12 protein sequences are pided into two types, the genome is located on 7 chromosomes, which more scattered than Arabidopsis. Protein secondary structure analysis shows that the amino acid sequences are composed of random curl as the main component. Gene expression is highest in both flesh and roots. And the results of this study provide an important basis for the further functional analysis of the Metacaspase gene family.
