    摘要:目的 本课题利用高脂饮食(high fat diet,HFD)建立小鼠肝损伤模型,研究紫甘薯花青素(purple sweet potato color,PSPC) 对高脂诱导的小鼠肝脏炎症反应的影响及分子机制。方法 将雄性ICR小鼠随机分成正常对照组、HFD组、HFD + PSPC组、PSPC组,以700mg /(kg •d)剂量灌喂PSPC,连续处理20周后处死,获取小鼠肝脏组织及血清、制备组织匀浆液。分别检测各组小鼠的肝脏湿重,血清中的丙氨酸氨基转移酶(ALT)活性,肝脏氧化应激状态以及炎症相关蛋白的表达。结果 HFD导致小鼠的体重、肝脏系数显著上升,血清ALT活性显著增强,肝脏组织中的活性氧(ROS)水平显著升高,超氧化物歧化酶(SOD)活力显著降低, NF-κB转录活性显著增强,iNOS和COX-2的表达水平显著升高。经PSPC处理,高脂诱导的小鼠肥胖症状得到有效缓解,小鼠肝脏的氧化应激指标恢复至正常水平,炎症反应显著减轻。结论 PSPC对肝脏有明显的保护作用,可能的机制是通过抑制炎症相关蛋白的表达,起到抗炎的功能。40231

    Purple Sweet Potato Color Attenuates Liver Inflammation Induced by High Fat Diet

    Abstract: Objective In this study, the liver injury mouse model induced by HFD (high fat diet) was used to explore the effect of PSPC (purple sweet potato color) on liver inflammation and the molecular mechanisms underlying this effect. Methods  Male ICR mice were randomly pided into 4 groups, termed as group control, group HFD, group HFD + PSPC, and group PSPC.Mice in group PSPC and group HFD + PSPC have daily oral administration of PSPC at doses of 700 mg/kg/day. After 20 weeks of treatment, mice were sacrificed. Liver tissue and serum were harvested to prepare the tissue homogenates. The wet weight of the liver, the activity of alanine aminotransferase (ALT), the oxidative stress state and the expression of inflammation related proteins were detected. Results The body weights, liver indexes, serum ALT activities and the hepatic levels of reactive oxygen species (ROS) were significantly increased, whereas the activities of superoxide dismutase (SOD) were markedly decreased in HFD-treated mice. Furthermore, the transcriptional activities of NF-κB, as well as the protein levels of inflammation related proteins (iNOS and COX-2), were notably elevated in the livers of HFD-treated mice. Interestingly, PSPC remarkably attenuated body weights, liver indexes, serum ALT activities, oxidative stress and inflammatory response in HFD-treated mice. Conclusion PSPC can effectively protect against HFD-induced liver injury by inhibiting inflammation in mice.
