摘要:本实验研究大孔吸附树脂对加拿大一枝黄花花序组织总多酚的分离和纯化作用,以及多酚提取物的体外抗氧化活性。采用70%乙醇提取加拿大一枝黄花花序组织粗多酚,以静态吸附和解吸附实验进行树脂类型的选择,在此基础上进行优选树脂类型对粗多酚的吸附和解吸附性能研究。结果显示,加拿大一枝黄花花序组织醇提取物中粗多酚的含量为110μg/mL,离子型吸附树脂NKA-9能有效吸附提取液中粗多酚成分,其优化的吸附工艺条件为树脂与粗多酚提取液的比例为1:25,上样液pH为8,上样流速为2mL/min;以乙醇为洗脱剂,优化的解吸工艺条件为解吸剂pH为9,乙醇体积分数为95%,洗脱流速为2mL/min。经树脂纯化后,多酚样品纯度为48.24%。粗多酚和纯化多酚的体外抗氧化活性研究表明:粗多酚对超氧阴离子自由基的抗氧化能力强于纯化物,对ABTS自由基、羟基自由基、DPPH自由基的清除能力低于纯化物。纯化多酚对不同自由基的清除活性大小依次是ABTS自由基>超氧阴离子自由基>羟基自由基>DPPH自由基,并具有较强的还原能力。本研究结果表明加拿大一枝黄花花序组织中有较高含量的多酚成分,并具有良好的抗氧化能力,为该类植物的有效利用提供数据参考。

Abstract:In this study, the total polyphenols in inflorescence tissue of Solidago canadensis L. were separated and purified by macroporous resin, and the antioxidant activities of the polyphenol extracts in vitro were studied. The crude polyphenols in the inflorescence of Solidago canadensis L. were extracted with 70% ethanol. The suitable resin type was selected on the base of static adsorption and desorption experiments, and the adsorption and desorption properties of the resin were studied. The results showed that the content of polyphenols in the alcohol extracts of the inflorescence tissue of Solidago canadensis
L. was 110μg/mL, and the ionic adsorbent resin NKA-9 could effectively adsorb the polyphenols in the extract. The optimum adsorption condition of macroporous resin NKA-9 was that the ratio of resin to sample is 1:25, the pH of sample solution is 8, and the sample upload rate is 2ml/min in dynamic adsorption experiment. The optimum desorption process was that the pH of desorption eluent is 9, the content of ethanol is 95% and the elution velocity was 2 ml/min in dynamic process. After purified by resin, the purity of polyphenol samples was 48.24%. Results of antioxidant activity of crude polyphenols and purified polyphenols showed that the antioxidant ability of crude polyphenol to superoxide anion radical is stronger than that of purified substance, the scavenging ability on ABTS radical, hydroxyl radical and DPPH radical was lower than that of purified product. The scavenging activities of purified polyphenols on different free radical were ABTS > superoxide anion radical > hydroxyl radical > DPPH. The purified substance also has stronger reducing power than crude extract. All of these results showed that the content of polyphenols in the inflorescence tissue of Solidago canadensis L. was high and the antioxidant activities were good, which can provide a useful reference for its effective utilization in the future.
