摘 要:通过改进微管固定,酶解及去除非特异性荧光步骤,初步建立棉花成熟组织微管免疫抗体染色方法。结果表明:该固定液可以非常好地文持细胞中微管结构的完整程度。此外, TritonX-100的加入在一定程度上减少了细胞骨架蛋白以外的蛋白成分。2%纤文素酶和1 %果胶酶比例的细胞壁水解酶混合液(含有0.5 mol•L-1甘露醇)处理材料,可以有效地降解细胞壁,同时又不影响细胞的原有形态。甲醇处理样品细胞可以除掉植物细胞之中的叶绿体的自行发生的荧光现象。此外,NaBH4处理可出去醛类物质引起的荧光。27863 毕业论文关键词:微管;免疫荧光抗体染色;棉花
Establish Immunfluorescence Antibody Staining of Microtubules Apical Tissue Levels on Cotton
Abstract: By improving the microtubules fixed and enzymatic processes as well as increasing the process of removing non-specific fluorescence to establish immunfluorescence antibody staining of chromosomes microtubules apical tissue levels on cotton.The results showed: improved fixative may well maintain the integrity of the cell structure of microtubules. In addition, addition of non-ionic detergent TritonX-100 in fixative can remove non-cytoskeletal proteins and soluble lipid components. A ratio of 1% cellulase plus 0.5% pectinase enzyme can fully digested cell wall, it will not destroy the cell morphology, 0.4 mol•L-1 mannitol may allow enzymatic solution osmotic potential and cell infiltration almost the same situation, it can be kept intact cell morphology. Methanol can be effectively removed in the plant cell chloroplast autofluorescence. In addition, NaBH4 treatment can effectively remove aldehydes triggered fluorescence.
