摘要:肉品的质量可以用许多技术指标来评定,但在实际的消费过程中,肉色对消费者来说是最容易判断的感观指标,肉色的好坏会直接影响着消费者的消费欲望,因此对生产者和消费者来说肉色是非常重要的肉质指标。在肉色形成过程中,肌红蛋白是影响肉色的主要蛋白,其结合配体与否、所结合配体的类型和铁离子的价态决定了肉色。但详细的分子调控机制及相关通路仍不清楚。近年来,国内外开展了大量关于肉色性状数量性状基因座鉴定的研究和肉色性状候选基因的挖掘,但目前能充足证明能影响肉色的关键基因还较少,可用于改良肉色性状的有效分子标记还十分匮乏。本研究在前期完成了高通量转录组测序研究工作基础之上,通过相关生物信息学分析与文献查阅,初步鉴定出影响猪肉色性状的关键候选基因 TFRC。在此基础上利用 Real-time PCR 技术对 TFRC 基因在具有不同肉色的猪骨骼组织,以及多种不同组织中的表达模式进行了检测。检测结果显示,TFRC基因在比目鱼肌、腰大肌等红肌内的表达水平较高,在股二头肌、半腱肌等白肌内的表达水平较低。TFRC 基因在红肌(比目鱼肌、腰大肌)、肝脏、肾脏、心脏等组织中表达水平较高。在白肌(股二头肌、半腱肌)、脾脏、肺、十二指肠、脂肪等组织中表达水平较低。本研究的结果可为深入开展猪 TFRC 基因对肉色性状形成的分子调控机制研究奠定理论基础,并为其今后作为影响猪肉色品质的候选基因的开发利用提供理论基础。30402 毕业论文关键词:猪;TFRC 基因;肉色;表达;
Pork color relevant candidate genes TFRC appraisal and tissueexpression pattern analysisAbstract: many technical indexes is available for meat quality evaluation, but in the actual consumption,color is the most easy cognitive sensory indicators to consumers,its quality directly affects the consumers'desire to buy, so color is a very important meat quality index for both producers and consumers.In theprocess of color formation, myoglobin is the main protein affecting the color,whether combination ofligand, the combination of the types of ligands and iron ion valence determines the color,but the specificmolecular regulation mechanism and related pathways are still unclear.In recent years,domestic andinternational scholars do a lot to study the identification of Color traits of quantitative trait loci and thecolor traits candidate genes,but there is few key genes which can be fully proved that influence thecolor,and effective molecular marker which can be used for improving color properties is very scarce. Inthe early study, high flux transcriptome sequencing research work has been completed.Throughbioinformatics analysis and literature review,preliminary identified the key candidate gene TFRC thataffects pork color traits.On this basis,use Real - time PCR technology to detection the differences of theexpression pattern of TFRC gene in the pig bone tissue of different color and different organizations.Theresults showed that,the expression level of TFRC genes in red muscle such as soleus and psoas ishigh,however the expression level in white muscle such as biceps femoris muscle and semitendinosus islow.Besides,the expression of TFRC genes in red muscles (soleus, psoas), liver, kidney, heart tissues are inhigher quantity,while in white muscle (biceps femoris muscle,semitendinosus), spleen, lung, duodenum,fat tissues the quantity is low.The results of this study can be used to lay the theoretical foundation forcarrying out the study of the regulatory mechanism of the pig TFRC gene in forming a nude traitsin-depth,and providing a theoretical basis for the development and utilization as the candidate gene thatinfluences the quality of pork color in future.
