摘要:硫酸软骨素是存在于动物体内的一类糖胺聚糖,因其具有降血脂、抗凝血、 抗肿瘤、促进冠状动脉循环和防止血管硬化等生物活性,广泛应用于临床医药、保健品食品、化妆品中。为了选育出硫酸软骨素高产量的菌株,以实验室提供的枯草芽孢杆菌为出发菌株,观察出发菌株形态特征,通过紫外诱变的方法对野生菌株进行优化,提高硫酸软骨素含量。实验得出,在距离紫外灯20cm,照射2min12s的诱变菌株UV3-9硫酸软骨素含量最高。在此基础上,对UV3-9的发酵条件进行了优化,获得了最优发酵条件:使用种龄为24h种子培养液,接种量为4%接种在装有50ml的250ml的三角烧瓶中,在转速为200rpm,温度为37℃的全温震荡培养箱中,培养48h。32838 毕业论文关键词: 硫酸软骨素;枯草芽孢杆菌;紫外诱变;发酵条件优化
Identification and mutation of chondroitin sulfate polysaccharides producing bacteria
Abstract:Chondroitin sulfate is present in animals of a class of glycosaminoglycan (GAG), because of its hypolipidemic, anticoagulant, anti tumor, promote coronary circulation and prevent hardening of the arteries and other biological activity, widely used in clinical medicine, health food and cosmetics. In order to select a strain of high yield of chondroitin sulfate, to provide laboratory of Bacillus subtilis as the starting strain observed starting strain morphological characteristics, through the method of UV mutagenesis of wild-type optimization, improve the content of chondroitin sulfate. The experiment, at a distance of 20cm UV light irradiation, the 2min12s mutant UV3-9 chondroitin sulfate content was the highest. Based, the fermentation conditions of UV3-9 for the optimization, won the optimal fermentation conditions: use age for 24h seed culture medium, inoculation amount 4%
inoculation with 50ml of 250ml Erlenmeyer flasks and in speed is 200rpm temperature temperature of 37 ℃shake culture box, cultivated for 48h.
