摘要:蜈蚣草是一种重金属砷的超积累植物,其地上部砷含量可达植物干重的2%。在自然环境中,五价砷 (As V)是土壤中砷的主要存在形式,同时也是蜈蚣草所吸收砷的主要来源。砷酸盐的化学结构与磷酸盐相似,蜈蚣草极有可能通过磷酸盐转运蛋白吸收土壤中的五价砷。在此,我们从蜈蚣草转录组数据库中分析整理了6个磷酸盐转运蛋白基因PvPht1;1-6,并对其进行了生物信息学分析和表达模式鉴定,结果表明:PvPht1;3和PvPht1; 6转录水平受As V诱导,在20μM As V处理条件下,表达量较野生型分别上调80、5倍。利用过量表达烟草材料,我们研究了PvPht1;3对As V吸收转运的影响。过量表达PvPht1;3的烟草株系中,地上部总砷含量较野生型提高了2-4倍,根系向地上部砷转运系数达到野生型的2-5倍。PvPht1;3的过量表达增强了植株对砷的吸收和从根系向地上部的转运。因此, PvPht1;3可能参与了蜈蚣草对砷的高效吸收转运过程。36610 毕业论文关键词:蜈蚣草 磷酸盐转运蛋白 As V PvPht1;3
Bioinformatics analysis and expression pattern identification of phosphate transporters in Peteris vittata and functional preliminary identification of PvPT3
Abstract:Peteris vittata is an arsenic hyperaccumulator which arsenic concentration in fronds is 2 percent of the dry fern。 Arsenate (As V) is the main arsenic species in soils and the main fern-available form of arsenic in the nature environment。 Arsenate was absorbed via the phosphate (Pi) transporters in soils for the similar chemical structure between Pi and As V。 Here, PvPht1; 1-6 are identified from the transcriptome database of Peteris vittata。 Employing qRT-PCR and bioinformatics analysis, expression pattern and bioinformatics characteristics of PvPht1; 1-6 have been determined。 The results showed that PvPht1;3 and PvPht1;6 transcripts were induced by AsV, which results in a significant up-regulation of the expression of PvPht1;3 by 80-fold and PvPht1; 6 by 5-fold under 20μM arsenic treatment。 Using PvPht1; 3 -overexpressing tobaccos, we investigated the impacts of overexpressing PvPht1; 3 on As V uptake and translocation。。 In this study, overexpression of PvPht1; 3 increased the total arsenic concentration of the shoots 2- to 4-fold compared with wild-type plants。 Arsenic transfer coefficient from root to shoot was 2- to 5-fold higher than wild-type plants。 It suggested that In a word, PvPht1; 3 overexpression enhanced arsenic uptake and root to shoot translation in transgenic tobacco。 PvPht1; 3 was involved in the arsenic efficient uptake and root to shoot translation in Peteris vittata。
