摘要:棉花是世界上最重要的纤文与油料作物,逆境胁迫影响了棉花的生长发育,严重影响了棉花的产量。丝裂原活化蛋白激酶(MAPK)级联途径是普遍存在于酵母、植物和动物等真核生物中的一类保守的信号转导通路。目前,越来越多的证据表明激素以及逆境可以激活MAPK级联系统,从而调控植物对外界的胁迫应答。现在棉花中已经分离到了一些MAPK及MAPKK,但棉花MAPKKK基因家族的相关研究还很少,并且至今尚未在棉花中发现一条完整的MAPKs信号通路。本研究通过生物信息学方法预测了到了89个棉花MAPKKK家族相关基因,并且从中分离鉴定了52个MAPKKK家族基因,再利用酵母双杂交技术,对棉花MAPKK与MAPKKK家族基因蛋白互作的关系进行了初步的研究。为进一步研究棉花MAPK级联途径的相关内容提供了更多的依据。37992 毕业论文关键词:棉花;MAPKKK基因家族;鉴定;蛋白互作
Protein Interactions Analysis of MAPKK and MAPKKK Family in Cotton MAPK Cascade Pathway
Abstract: Cotton is the world’s most important natural textile fiber and oilseed crop. Stressors severely affect growth and development and limit crop productivity and quality. Mitogen-activated protein kinase (MAPK) cascades are highly conserved signaling modules found in all eukaryotes, including fungi,plants and animals which play a important role in stress responses of plant.At present, more and more evidence indicate that hormones and adversity can activate the MAPK cascade system, which regulates the stress response plants to the outside world.Some MAPKs and MAPKKs have been isolated from cotton, but there are few studies on the MAPKKK gene family, and a complete MAPKs signaling pathway has not yet been found in cotton.In this study, 89 MAPKKK genes were predicted by bioinformatics, and in which 52 MAPKKK genes were isolated and identified. And protein interactions between MAPKK and MAPKKK gene was preliminary studied by yeast two-hybrid technique.Which provided more basis for further study in cotton MAPK cascade pathway.

棉花MAPK级联途径中MAPKK与MAPKKK家族基因蛋白互作分析
引言:棉花是世界上最重要的纤文与油料作物,产量极为重要,严重的逆境胁迫影响了棉花的生长发育。所以从作物遗传育种方面研究棉花的抗逆境胁迫具有重要意义。在植物的生命周期中,不利的环境胁迫可以通过信号传导途径将信号传递至植物细胞内,从而调控基因表达对逆境胁迫作出适应性反应,减少逆境对植物的不良影响。丝裂原活化蛋白激酶(MAPK)级联途径是普遍存在于酵母、动物和植物等真核生物中的一类保守的信号转导通路,MAPK级联途径在植物中的研究成果越来越多,不断积累的成果表明MAPK级联途径及其相关基因,在植物的生长发育、激素、细胞分裂调控以及生物和非生物胁迫等过程中具有重要作用。
