摘要:番茄(2n=24)基因组大小约950Mb,存在着较丰富的近缘种质材料。采用双色FISH技术对两个BAC克隆探针在野生型和茄科近缘种的有丝分裂中期染色体上进行了荧光定位核型分析。用来自番茄的45S rDNA克隆pTa71与番茄及其近缘种进行的原位杂交,定位结果显示在近端着丝粒染色体末端的球形随体上有较强的杂交信号,分析出茄科及其近缘种基因组中只有随体含有45S rDNA单位的遗传编码区,番茄只有一对45S rDNA位点,杂交信号显示随体极易断裂。用番茄5S rDNA,该克隆为pCT4.2进行的荧光原位杂交在核仁组织区显示出杂交信号,检测出番茄只有一对5S rDNA位点。文中基于野生番茄与番茄近缘种的45S rDNA&5S rDNA-FISH核型分析图,讨论了它们在番茄与番茄近缘种的基因组中保守性和不同的进化趋势。40449 毕业论文关键词:番茄;FISH;rDNA;核型分析
Karyotype Comparative Analysis of rDNA Distribution in Metaphase Chromosomes of Tamato Species
Abstract: Tomato genome size is about 950Mb, and there is a rich relatives germplasm in solanaceae plants. The karyotype analyses about the wild tomatoes and its relatives germplasm were finished employing dual-color FISH technology with two BAC clones probes to located on the mitotic metaphase chromosomes. Fistly, when 45SrDNA from tomatoes pTa71 clone was used as probes in the FISH experiments, the result show that there is strong hybridization signal only in the satellite body to the end of the acrocentric chromosome, and only the chromosome satellite contains the coding regions encoding 45SrDNA in plants of solanaceae and its relatives germplasm, and there are a pair of 45S rDNA loci in tomato, and the results show, too, that the satellite is easy to fracture. Secondly, when another probe, the 5S rDNA from pCT4.2 of tomato, was used in FISH, it was found that the sites, only in Nucleolus Organizer Region with the strong signal, doubles. In this study, the conservative and evolution trend of genomes of tomatoes and its relatives is discussed based on karyotype analysis from 45S rDNA&5S rDNA-FISH.
