摘要:本研究分析了病原菌细胞壁、IPTG、乳糖、葡聚糖对木霉T-Aloe产β-1,3-葡聚糖酶的诱导作用。SDS-PAGE蛋白电泳结果表明,木霉T-Aloe产生的葡聚糖酶为80KDa左右。酶活性分析表明,诱导24h时,在未加入任何诱导剂的情况下测得的酶活性较高,最高酶活为1.195 U/(ml•min);当诱导72h时,除加入IPTG和乳糖为诱导剂测得的酶活有所下降,其余各组的酶活性均有所升高,其中加入病原菌细胞壁的的两组酶活增加较多,分别提高0.116U/(ml•min)、0.148U/(ml•min)。40448 毕业论文关键词:功能分析;木霉T-Aloe;葡聚糖酶
Function analysis of β -1,3-glucanase from Trichoderma T-Aloe
Abstract:The inductions of β-1,3-glucanase by pathogen cell wall, IPTG, lactose, glucanase were analyzed. The protein of 80KDa expressed was determined by SDS-PAGE protein electrophoresis. Enzyme activity analysis showed that the highest activity was 1.195 U / (ml•min) when being added any inducer after culturing 24h. Enzyme activity of other tests all increased in addition to IPTG and lactose inducers after culturing 72h. The increase of activity was much higher when pathogenic bacteria cell wall was added. These were 0.116 U / (ml•min)、0.148 U / (ml•min).
