摘要目的:提取并检测苦参总碱(Alkaloids of Sophora Flavescens, ASF)和淫羊藿总黄酮(total flavone of Herb Epimedii,TFE)对人脐带静脉内皮细胞(human umbilical vein endothelial cell, HUVEC)增殖的影响。方法:水煎煮法提取苦参总碱,醇提取法提取淫羊藿总黄酮,以人脐静脉内皮细胞为模型,采用MTT法检测这两者对人脐静脉内皮细胞增殖的影响。结果:苦参总碱和淫羊藿总黄酮提取得率分别为77.8%和1.65%。MTT实验结果表明苦参总碱和淫羊藿总黄酮都对HUVEC有一定的抑制作用。4mg/ml苦参总碱作用HUVEC细胞24、48、72 h,对细胞生长的抑制率分别为44.2%,59.6%,77.6%。100μg/ml淫羊藿总黄酮作用HUVEC细胞24、48、72 h,对细胞生长的抑制率分别为56.5%,70.8%,76.5%。结论:苦参总碱和淫羊藿总黄酮均能明显抑制人脐带静脉内皮细胞的生长,且其抑制的效果与剂量和作用时间有关。44201
Objective: To extract and study the inhibitive effect of alkaloids of Sophora flavescens (ASF) and total flavones of Herb epimedii (TFE) on human umbilical vein endothelial cell (HUVEC) proliferation. Method: Alkaloids was extracted from Sophora flavescens with water decocting. Well the total flavones was extracted from Herb Epimedii with alcohol decocting. Cell growth inhibitory effect of ASF and TFE on HUVEC cells was measured by MTT assay. Results: ASF displayed growth inhibitory activity against HUVEC cells with inhibition ratio of 44.2%, 59.6% and 77.6% for 24, 48, 72 hours in the 4mg/ml ASF treated group. And the TFE showed the same effect, its inhibition ratio for 24, 48, 72 hours in the 100μg/ml TFE treated group goes to be 56.5%, 70.8% and 76.5%.
Conclusion: The alkaloids of Sophora flavescens and total flavones of Herb epimedii had obvious inhibition on human umbilical vein endothelial cell, and the effect was time- and concentration-dependent.

1引言
我国药用植物资源丰富,现有记载的就有11,146种。目前我国药用植物资源的开发和利用己逐步形成多层次、多方位和多学科研究的特点[1] 。中药是祖国医学用来防治疾病的有力武器。人们发现中药在治疗恶性肿瘤方面,无论是在减轻临床症状、提高生存质量、防止复发转移、延长生存期,还是在与放化疗配合、增效减毒等方面都有很好的效果[2]。
随着科学技术的进步,人们对于中药的提取开始向简便、科学、高效、可控的方向发展,传统的溶剂提取法对于中药有效成分的提取都有一定的局限。广泛尝试运用新的方法来优化苦参的有效药用成分的提取工艺,对提高资源利用率、保护环境有重要意义。目前使用的传统方法主要是溶剂提取法,随着工业技术的发展,一些高新的工业技术被应用到传统中药的生产中来,如微波提取法、胶束提取、离子树脂交换法、树脂吸附法、动态逆流提取、超临界CO2萃取法、酶提取法、超声提取法、减压沸腾提取、半仿生提取法等。
