摘要:本课题采用微卫星标记技术,对铜陵、当涂、安庆、太湖、高邮和靖江6个不同群体鳜的96个个体的遗传多样性进行了初步分析。结果表明,经筛选的10对引物在6个鳜群体中的平均等位基因数(Na)为2.2000-2.5000,期 望 杂 合 度(He)为0.6489-0.7289,多 态 信 息 含 量(PIC)为0.5614-0.6409(PIC>0.5),遗 传 分 化 系 数(Fst)为0.0606(0.05<Fst<0.15),平 均 基 因 流(Nm)为4.0151(Nm>1),揭示了6个 鳜群 体的遗传多 样性 较高,群体间 遗 传分 化水平中 等,存在着一 定的基 因交流。检验表明,除高邮和靖江群 体外,其他群 体均不同程 度地偏 离Hardy-Weinberg平 衡,主要表现为杂 合 子缺失。构建的UPGMA聚类图显示,铜陵、安庆、太湖、高邮和靖江群 体聚 为一类,当涂群 体聚 为另一类。26590 毕业论文关键词:鳜;不同群体;微卫星标记;遗传多样性
Analysis of genetic persity in six populations of Siniperca Chuatsi based on SSR
Abstract:Ten microsatellite markers were used to analyze the genetic structure in six populations of Siniperca Chuatsi. In each population, the average of allele value was from 2.2000 to 2.5000, the average expected heterozygosity (He) value was from 0.6489 to 0.7289 and the average polymorphic information content (PIC) value was from 0.5614 to 0.6409. The genetic differentiation coefficient among the population (Fst) was 0.0606, and the gene flow (Nm) was 4.0151. These genetic statistics indicated a richer genetic persity in six populations, there were middle genetic differentiation among these six populations and strong gene exchange among them. Same loci were found to be deviated from Hardy-Weinberg Equilibrium in each population, and it indicated that the deficiency of heterozygote existed in six populations. The UPGMA clustering showed that dangtu population formed an independent cluster,the other five populations were another cluster.
